BPX-201 + AP1903 is a combination immunotherapy developed by Bellicum Pharmaceuticals for the treatment of metastatic castration-resistant prostate cancer (mCRPC). BPX-201 is an autologous dendritic cell vaccine engineered with the proprietary DeCIDe "on switch" and trained to target prostate cancer cells, specifically those expressing prostate-specific membrane antigen (PSMA). The dendritic cells are transduced with an adenoviral vector encoding inducible MyD88/CD40 signaling elements, enhancing their ability to stimulate cytotoxic T lymphocytes. After administration of the vaccine, patients receive AP1903 (also known as rimiducid), a small molecule dimerizer that activates the engineered signaling pathway in vivo. This activation is designed to occur after dendritic cells have migrated to lymph nodes, aiming for a more potent and durable antigen-specific T cell immune response. The combination was evaluated in phase I clinical trials but development was discontinued following company wind-down[1][2][5][7].
