BPX-601 + rimiducid is an investigational combination therapy in which **BPX-601** is an autologous chimeric antigen receptor T cell (CAR-T) product engineered to target **prostate stem cell antigen (PSCA)**. It incorporates an inducible MyD88/CD40 (iMC) costimulatory ON-switch, which allows T cell activation and expansion upon administration of **rimiducid**, a small-molecule dimerizer. Unlike standard CAR-T cells, BPX-601 GoCAR-T cells can be pharmacologically activated or modulated in vivo using rimiducid, providing external control over CAR-T cell activity, persistence, and potential toxicity. The combination has been studied in phase 1 trials for metastatic pancreatic ductal adenocarcinoma (mPDAC) and metastatic castration-resistant prostate cancer (mCRPC). Clinical benefits, such as PSA declines and tumor reduction, have been reported in mCRPC. The therapy was generally well tolerated but dose-limiting toxicities and treatment-related deaths occurred at higher doses in mCRPC[1][2][3][5]. BPX-601 and rimiducid are being developed by **Bellicum Pharmaceuticals**.
