**BPX-603 + rimiducid** is an investigational combination therapy in which patients receive BPX-603, a dual-switch, HER2-targeted autologous chimeric antigen receptor (CAR)-T cell product, followed by rimiducid, a small molecule used to activate the signaling of the engineered T cells. BPX-603 incorporates both an inducible MyD88/CD40 (iMC) activation switch and a CaspaCIDe safety switch, allowing physicians to control CAR-T cell activity and mitigate toxicity. Rimiducid is administered intravenously to activate the iMC switch post-infusion, enhancing T cell persistence and function. The combination was developed by Bellicum Pharmaceuticals for treatment of various HER2-positive solid tumors (e.g., breast, gastric, ovarian, colorectal, endometrial cancers). Clinical trials were terminated in early phases due to safety concerns, notably severe cytokine release syndrome.
