Overview

Brentuximab vedotin + dacarbazine is a combination of an antibody-drug conjugate (brentuximab vedotin) and an alkylating agent (dacarbazine). **Brentuximab vedotin** targets CD30, a protein expressed on classical Hodgkin lymphoma cells, delivering a potent microtubule-disrupting agent, monomethyl auristatin E (MMAE), into CD30-expressing cells via a protease-cleavable linker. **Dacarbazine** is a cytotoxic chemotherapy that acts as an alkylating agent, leading to DNA damage and apoptosis in rapidly dividing cells. This combination has been investigated mainly for the treatment of classical Hodgkin lymphoma, particularly in combination with other agents, such as doxorubicin and vinblastine, but the direct two-drug combination has not been reported as a standalone approved regimen. The mechanism of the combination leverages the targeted cytotoxicity of brentuximab vedotin with the nonspecific DNA-damaging effect of dacarbazine.
