Brentuximab vedotin + gemcitabine is a combination regimen of an antibody-drug conjugate (brentuximab vedotin) and a small molecule antimetabolite (gemcitabine), both of which are used to treat certain cancers. Brentuximab vedotin consists of a monoclonal antibody targeting CD30 conjugated to the cytotoxic microtubule-disrupting agent monomethyl auristatin E (MMAE), designed for selective delivery to CD30-positive tumor cells, leading to microtubule disruption, cell cycle arrest, and apoptosis. Gemcitabine is a deoxycytidine analog and a nucleoside metabolic inhibitor that is phosphorylated in cells to active metabolites; these incorporate into DNA to inhibit synthesis and lead to apoptotic cell death. This combination has been investigated for treating various lymphomas, especially in patients with relapsed or refractory disease. The combination exploits distinct, complementary anticancer mechanisms—targeted antibody-directed cytotoxicity and nucleoside-based inhibition of DNA replication—enhancing efficacy against malignant cells.
