Overview

**Brentuximab vedotin + pembrolizumab + doxorubicin + dacarbazine** is an investigational combination regimen for relapsed/refractory classical Hodgkin lymphoma (HL), particularly high-risk cases. Brentuximab vedotin is an **antibody-drug conjugate** targeting **CD30** with monomethyl auristatin E payload to induce cell death, pembrolizumab is a **PD-1 inhibitor** monoclonal antibody blocking immune checkpoints, doxorubicin is an **anthracycline** that intercalates DNA and inhibits topoisomerase II, and dacarbazine is an **alkylating agent** that methylates DNA causing crosslinks. Developed through clinical trials evaluating multi-agent approaches in heavily pretreated patients, it aims to achieve complete metabolic remission as a bridge to autologous stem cell transplantation (ASCT), showing promising efficacy in retrospective and phase II studies for R/R HL.
