**BRII-179 + elebsiran** is an investigational combination therapy for chronic hepatitis B virus (HBV) infection. BRII-179 is a novel recombinant protein-based therapeutic vaccine expressing Pre-S1, Pre-S2, and S HBV surface antigens to induce broad B-cell and T-cell immunity, aimed at restoring or enhancing the host immune response against HBV. Elebsiran (previously known as BRII-835) is a small interfering RNA (siRNA)-based therapy designed to selectively silence HBV transcripts, reducing production of hepatitis B surface antigen (HBsAg) and thereby promoting viral clearance. The combination leverages immunostimulation from the vaccine and direct viral suppression by siRNA. This strategy aims to improve functional cure rates for chronic HBV, as indicated by HBsAg loss and seroconversion. The drugs are being developed by Brii Biosciences and are undergoing multi-phase clinical evaluation, including Phase 2 studies assessing immunological and virological endpoints in patients with chronic hepatitis B[1][3][4][5][7][10][11].
