Overview

BRII-179 (also known as VBI-2601) combined with interferon-alpha (specifically pegylated interferon-alpha, PEG-IFNα) is an investigational therapeutic regimen for chronic hepatitis B infection. BRII-179 is a novel recombinant protein-based immunotherapeutic vaccine that expresses the Pre-S1, Pre-S2, and S surface antigens of hepatitis B virus (HBV). Its mechanism of action involves priming and boosting HBV-specific B-cell and T-cell immune responses to enhance seroconversion rates and promote HBsAg loss. When used in combination with PEG-IFNα—a cytokine with antiviral and immunomodulatory properties—this regimen aims to achieve a functional cure by inducing robust humoral and cellular immunity against HBV. Clinical studies have shown that the combination increases rates of HBsAg loss compared to standard therapy alone, with a favorable safety profile[1][2][4][5].
