**Brimarafenib + panitumumab** is an investigational combination therapy under clinical evaluation for advanced or metastatic colorectal cancer. Brimarafenib (BGB-3245) is a selective small molecule inhibitor of RAF kinases, including mutant BRAF (such as BRAF V600E), designed to block MAPK pathway signaling. Panitumumab is a fully human monoclonal antibody (IgG2) targeting the epidermal growth factor receptor (EGFR), blocking ligand-induced EGFR activation and downstream signaling. The combination is being studied to overcome resistance seen with single-agent RAF or EGFR inhibition, leveraging dual inhibition to provide greater efficacy in BRAF-mutant colorectal cancers by simultaneously targeting mutated BRAF-driven and EGFR-driven signaling pathways. This approach is expected to reduce or delay tumor escape mechanisms that limit the efficacy of single-agent therapies[1][7][5][3][8].
