This combination comprises **brivanib alaninate** (BMS-582664), an oral prodrug that inhibits vascular endothelial growth factor receptor (VEGFR) and fibroblast growth factor receptor (FGFR) tyrosine kinases; **5-fluorouracil** (5-FU), a pyrimidine analogue antimetabolite that inhibits thymidylate synthase; **leucovorin**, which enhances the effects of 5-FU by stabilizing the 5-FU-thymidylate synthase complex; and **irinotecan**, a topoisomerase I inhibitor causing DNA damage. The combination targets angiogenesis, tumor cell proliferation, and DNA replication in cancer cells. Brivanib is under development primarily by Bristol Myers Squibb for advanced/metastatic colorectal cancer and other gastrointestinal malignancies[2][3].
