This is a combination chemotherapy regimen consisting of four agents: - **Bromodeoxyuridine** (BrdU): A thymidine analog that incorporates into DNA during replication, making cells more sensitive to DNA-damaging agents and radiation. It acts as a radiosensitizer and antimetabolite. - **Lomustine** (also known as CCNU): An alkylating agent that cross-links DNA, inhibiting cancer cell replication and leading to cell death[1][2][3][4]. - **Procarbazine**: An alkylating agent that inhibits DNA, RNA, and protein synthesis by causing breaks in the DNA strands. - **Vincristine**: A vinca alkaloid that inhibits microtubule formation in mitosis, arresting cell division. This combination is designed for synergistic cytotoxic effects against rapidly dividing tumor cells. While the three-drug regimen of procarbazine, lomustine (CCNU), and vincristine—known as PCV—is well established for treating brain tumors such as gliomas[6][8][10], the addition of bromodeoxyuridine is less common but may be used experimentally or in clinical trials to enhance treatment efficacy through radiosensitization.
