Overview

**Budigalimab + rovalpituzumab tesirine** is a combination therapy investigated for advanced solid tumors, especially small cell lung cancer (SCLC)[3][1][7][5][9]. Budigalimab is a humanized IgG1 monoclonal antibody that targets the programmed cell death 1 (PD-1) receptor on T-cells, functioning as an immune checkpoint inhibitor by blocking PD-1/PD-L1 interactions and reactivating exhausted T-cells for anti-tumor effects[2][3][4][8]. Rovalpituzumab tesirine is an antibody-drug conjugate (ADC) targeting delta-like protein 3 (DLL3), a protein highly expressed on SCLC cells. Rovalpituzumab tesirine delivers a cytotoxic payload directly to DLL3-expressing tumor cells, resulting in cell death. The combination was tested in the NCT03000257 phase I clinical trial, where patients received intravenous budigalimab and Rova-T infusions as separate agents for synergistic antitumor activity. While the combination demonstrated tolerability and clinical activity in SCLC, development of rovalpituzumab tesirine has since been discontinued due to evolving company strategies[3].
