Bumetanide + mebendazole is a small molecule combination therapy under development by Neurochlore and B&A Oncomedical for the treatment of glioblastoma and other brain tumors. The combination utilizes bumetanide to inhibit the Na-K-2Cl cotransporter 1 (NKCC1), which regulates intracellular chloride levels and is associated with glioma cell migration and peritumoral edema, alongside mebendazole, a benzimidazole anthelmintic that disrupts microtubule formation by binding to tubulin. This dual approach is intended to inhibit tumor growth and invasion more effectively than either agent alone. The project is currently in preclinical development, supported by data from tumoroid models and murine brain slices, and has been used in a compassionate pilot trial.
