BKM120 (buparlisib) + cisplatin is a combination therapy that pairs a pan-class I PI3K inhibitor with a platinum-based chemotherapy agent. BKM120 works by blocking the phosphatidylinositol-3-kinase (PI3K) signaling pathway, which plays a prominent role in cancer development. When combined with cisplatin, this therapy has shown potential synergistic effects in inhibiting cancer cell growth, particularly in colorectal cancer cells. Studies have demonstrated that inhibition of PI3K/AKT signaling using BKM120 can reduce the metabolic activity of cancer cells in a concentration and time-dependent manner, inducing G2/M cell cycle arrest and apoptosis-mediated cell death. The combination has also been shown to inhibit cancer invasion and remarkably sensitize cancer cells to cisplatin.
