**Burixafor + propranolol** is an investigational combination therapy consisting of **burixafor (GPC-100)**, a potent and selective small molecule antagonist of the chemokine receptor **CXCR4**, and **propranolol**, a non-selective beta-adrenergic receptor antagonist (beta-blocker). Preclinical studies and ongoing phase 2 clinical trials have demonstrated that this combination significantly enhances the mobilization of hematopoietic stem cells (HSCs) and white blood cells compared to standard mobilization regimens. The rationale stems from the functional crosstalk between CXCR4 and β2-adrenergic receptor (β2AR); their simultaneous blockade (by burixafor and propranolol, respectively) leads to augmented mobilization of HSCs and immune cells. This regimen is being evaluated in multiple myeloma and other hematologic conditions requiring autologous stem cell transplantation, and is particularly valuable for patients who poorly tolerate granulocyte colony-stimulating factor (G-CSF) or in cases of inadequate stem cell mobilization.
