BVAC-C + durvalumab is a combination immunotherapy regimen under investigation for the treatment of recurrent or metastatic human papillomavirus (HPV) 16/18-positive cervical cancer. - **BVAC-C** is a novel B cell- and monocyte-based therapeutic cancer vaccine transfected with recombinant E6/E7 genes from HPV types 16 and 18. It aims to stimulate an anti-tumor immune response by presenting HPV oncoproteins to the immune system, thereby activating cytotoxic T cells against HPV-infected tumor cells. - **Durvalumab** is a fully human monoclonal antibody that targets programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80 on T cells. This releases the "brakes" on the immune system, enhancing anti-tumor immunity. The rationale for combining these agents is to synergistically enhance anti-tumor responses by both directly targeting viral antigens in tumor cells (via BVAC-C) and overcoming tumor-induced immunosuppression (via PD-L1 blockade with durvalumab). Clinical trials have shown promising efficacy and manageable safety in patients who have progressed after platinum-based chemotherapy[1][2][3][4][5].
