C-192, chemically known as calix[4]arene tetrakis-methylene-bis-phosphonic acid, is a small molecule research compound designed as a potent inhibitor of fibrin polymerization for potential antithrombotic applications. It is a calixarene derivative featuring four methylenebisphosphonic acid groups at the macrocyclic upper rim. The compound's mechanism of action involves blocking the fibrin polymerization site 'A' (specifically the Aα17-19 region), which prevents the initial 'knob-hole' interaction required for protofibril formation during blood coagulation. In preclinical studies, C-192 demonstrated significant potency in inhibiting fibrin polymerization and was shown to prolong both prothrombin time (PT) and activated partial thromboplastin time (aPTT) in human blood plasma. While it serves as a promising platform for the development of a new class of antithrombotic agents, it remains in the preclinical research stage with no specific commercial developer or brand name currently associated with it.
