Overview

C1-R212 + C2-R212 is a combination therapeutic being developed by Rejuvenate Bio. The program, often referred to as R212, utilizes a dual-gene therapy approach delivered via an adeno-associated virus (AAV) vector. It is designed to treat chronic kidney disease (CKD) by delivering two distinct therapeutic genes: one encoding for Fibroblast Growth Factor 21 (FGF21) and the other for the soluble extracellular domain of alpha-Klotho (sKlotho). FGF21 is a metabolic regulator that improves insulin sensitivity and reduces inflammation, while Klotho is a potent anti-aging protein that protects renal function and prevents vascular calcification. By co-expressing these factors, the therapy aims to address the complex pathophysiology of CKD, including fibrosis and metabolic dysfunction, potentially slowing or reversing disease progression.
