This combination therapy regimen consists of cabozantinib, a multi-targeted small molecule tyrosine kinase inhibitor, and standard androgen ablation therapy (ADT). Cabozantinib targets several receptor tyrosine kinases involved in tumor growth, angiogenesis, and bone metastasis, specifically MET, VEGFR2, and RET. Androgen ablation therapy, which can be achieved through luteinizing hormone-releasing hormone (LHRH) agonists, LHRH antagonists, or surgical castration, serves to deplete circulating androgens that drive prostate cancer cell proliferation. This specific combination was evaluated in a Phase 2 clinical trial led by the M.D. Anderson Cancer Center for patients with newly diagnosed hormone-naïve metastatic prostate cancer, aiming to improve progression-free survival by simultaneously targeting the androgen receptor pathway and alternative growth signaling pathways.
