Overview

Cabozantinib + atezolizumab is a combination therapy consisting of cabozantinib, an oral small molecule tyrosine kinase inhibitor, and atezolizumab, a monoclonal antibody immune checkpoint inhibitor targeting PD-L1. This combination is being investigated for the treatment of various advanced solid tumors, including metastatic castration-resistant prostate cancer (mCRPC), renal cell carcinoma (RCC), hepatocellular carcinoma (HCC), and non-small cell lung cancer (NSCLC). Cabozantinib inhibits multiple receptor tyrosine kinases involved in tumor growth and angiogenesis—specifically MET, VEGFRs, and AXL—while also modulating the tumor microenvironment to promote an immune-permissive state. Atezolizumab blocks PD-L1 on tumor cells and immune cells to enhance T-cell–mediated antitumor responses. The combination has demonstrated synergistic clinical activity with improved progression-free survival compared to standard therapies in mCRPC and other cancers[1][3][5][6]. The safety profile is consistent with known effects of each agent.
