Overview

Cabozantinib + ipilimumab + nivolumab is a triplet combination therapy that combines a tyrosine kinase inhibitor (cabozantinib) with two immune checkpoint inhibitors (ipilimumab and nivolumab). This combination has been studied primarily for advanced renal cell carcinoma (RCC) and other genitourinary tumors. The triplet therapy works through complementary mechanisms: cabozantinib inhibits multiple receptor tyrosine kinases involved in tumor growth and angiogenesis (including MET, VEGFR, and the TAM family of kinases), while nivolumab (anti-PD-1) and ipilimumab (anti-CTLA-4) promote antitumor immune responses through different but complementary immune checkpoint inhibition pathways.
