Cabozantinib + trastuzumab is an investigational combination therapy consisting of two agents with distinct mechanisms of action. Cabozantinib is a small molecule tyrosine kinase inhibitor that targets multiple kinases involved in tumor growth, angiogenesis, and metastasis, including MET, VEGFR2, AXL, ROS1, TYRO3, MER, KIT, TRKB, FLT3, and TIE2. Trastuzumab is a recombinant humanized monoclonal antibody that binds to the extracellular domain of the HER2 receptor on tumor cells. The combination has been studied primarily in patients with HER2-positive breast cancer brain metastases to evaluate efficacy and tolerability. Cabozantinib inhibits pathways critical for tumor vascularization and invasiveness while trastuzumab blocks HER2-mediated signaling and induces immune-mediated cytotoxicity against HER2-overexpressing cells[1][4][6][8].
