Overview

Cadonilimab + pulocimab is a combination of two investigational monoclonal antibodies developed by Akeso for the treatment of advanced solid tumors, especially cancers resistant to previous immunotherapy. **Cadonilimab** is a first-in-class symmetric tetravalent bispecific antibody that concurrently targets PD-1 (Programmed cell death protein 1) and CTLA-4 (Cytotoxic T-lymphocyte-associated protein 4) immune checkpoints on T cells. This dual blockade is designed to stimulate T cell activation and overcome tumor-mediated immune suppression by blocking these key inhibitory pathways. Its Fc–null design minimizes unwanted immune cell activation. **Pulocimab** is a monoclonal antibody that targets VEGFR-2 (Vascular endothelial growth factor receptor 2), inhibiting angiogenesis and disrupting the tumor’s blood supply. The combination synergistically targets immune escape and angiogenesis, offering a potentially effective option for patients with non-small cell lung cancer (NSCLC), gastric, gastroesophageal junction (G/GEJ) cancers, hepatocellular carcinoma, and potentially other solid tumors after failure of immunotherapy[1][4][13]. Clinical trials have demonstrated promising efficacy and safety in immunotherapy (IO)-resistant NSCLC and gastric/GEJ cancer[1][4].
