CAIX.CD30(ICD) CAR-T is an experimental next-generation chimeric antigen receptor (CAR) T-cell therapy designed to treat solid tumors, specifically targeting Carbonic Anhydrase IX (CAIX). Unlike conventional second-generation CARs that rely solely on antigen-triggered costimulation (e.g., CD28 or 4-1BB), this construct incorporates the intracellular domain (ICD) of CD30. The CD30 ICD provides autonomous, ligand-independent costimulatory signaling by constitutively activating the NF-κB pathway. This mechanism enhances T-cell proliferation, cytokine secretion (IFN-γ, IL-2), and persistence while reducing exhaustion markers (PD-1, LAG-3, Tim-3). Preclinical studies demonstrate superior antitumor activity in orthotopic renal cell carcinoma and pulmonary metastasis models.
