GK664S (MC2-01) is a topical fixed-dose combination of calcipotriol and betamethasone dipropionate, formulated using the proprietary Polyaphron Dispersion (PAD) Technology. Developed by MC2 Therapeutics and licensed to Maruho for the Japanese market, the PAD technology enables the creation of a stable, aqueous-based cream that aims to improve skin penetration and patient convenience compared to traditional ointment-based treatments. Calcipotriol is a vitamin D3 analog that binds to the vitamin D receptor (VDR), regulating keratinocyte proliferation and differentiation. Betamethasone dipropionate is a potent corticosteroid that acts as a glucocorticoid receptor agonist, providing anti-inflammatory, antipruritic, and immunosuppressive effects. The product is marketed as Wynzora in the United States and Europe and is currently in Phase III clinical trials in Japan for the treatment of plaque psoriasis (psoriasis vulgaris).
