calcipotriol + betamethasone dipropionate (LEO Pharma)

Daivobet/Dovobet Gel is a topical fixed-dose combination therapy developed by LEO Pharma for the treatment of plaque psoriasis (psoriasis vulgaris). It combines calcipotriol, a vitamin D3 analogue, and betamethasone dipropionate, a potent corticosteroid. Calcipotriol acts by binding to the vitamin D receptor (VDR) to inhibit keratinocyte proliferation and promote normal differentiation, addressing the underlying cellular dysfunction in psoriasis. Betamethasone dipropionate acts on the glucocorticoid receptor to provide anti-inflammatory, antipruritic, and immunosuppressive effects. The gel formulation was designed for once-daily application and received its first major approvals in 2008. It is marketed as Taclonex in the United States and has been evaluated in large-scale Phase 4 studies like PSO-TOP to optimize outpatient care.
