Calcipotriol + etanercept is a combination therapy used off-label for the treatment of moderate to severe plaque psoriasis and psoriatic arthritis, particularly in patients who have an inadequate response to etanercept monotherapy. Calcipotriol is a synthetic vitamin D3 analogue that acts by binding to the vitamin D receptor, modulating keratinocyte proliferation and differentiation, and exerting anti-inflammatory effects. Etanercept is a recombinant fusion protein that functions as a tumor necrosis factor (TNF) inhibitor by binding TNF-alpha and preventing its interaction with cell surface receptors, thereby reducing inflammation. The combination aims to enhance therapeutic efficacy while minimizing cumulative toxicity or side effects associated with higher doses of either agent alone[1][3][4].
