Calcipotriol + LEO80122 is a combination topical drug developed for the treatment of psoriasis vulgaris. Calcipotriol (also known as calcipotriene) is a synthetic vitamin D3 analogue that acts primarily by binding to the vitamin D receptor (VDR) on keratinocytes and immune cells in the skin. This interaction modulates gene expression involved in cell differentiation and proliferation, leading to reduced abnormal growth of skin cells characteristic of psoriasis. The precise mechanism involves VDR agonism with downstream effects on T-cell function and inflammatory cytokine production[2][6][8]. The combination was investigated under code names such as LEO 19123 for its efficacy in treating plaque psoriasis but development appears to have been discontinued after phase 2 trials[4].
