calcitriol + dexamethasone

Calcitriol + dexamethasone is an investigational combination of two pharmaceutical agents. Calcitriol is the active form of vitamin D3 (1,25-dihydroxycholecalciferol), which increases calcium absorption and regulates parathyroid hormone levels. Dexamethasone is a synthetic glucocorticoid corticosteroid with potent anti-inflammatory and immunosuppressive properties. Preclinical studies have suggested potential antitumor synergy between these agents, particularly in prostate cancer models. The combination has been evaluated in clinical trials for castration-resistant prostate cancer, where high-dose intravenous calcitriol was administered alongside oral dexamethasone; however, no significant clinical or PSA responses were observed in phase 2 studies[1][4]. The regimen was generally well tolerated but did not demonstrate sufficient efficacy to warrant further development for this indication.
