Calcitriol + docetaxel is a combination therapy investigated primarily for the treatment of metastatic androgen-independent prostate cancer (AIPC). Calcitriol is the hormonally active form of vitamin D and acts as a natural ligand for the vitamin D receptor, exerting antineoplastic effects through induction of apoptosis, cell cycle arrest, differentiation, inhibition of proliferation, invasion, and angiogenesis. Docetaxel is a taxoid antineoplastic agent that promotes microtubule assembly and stabilizes them by preventing depolymerization; this disrupts normal microtubule dynamics essential for mitosis and interphase cellular functions. The combination has shown promising efficacy in phase II clinical trials with high PSA response rates and manageable toxicity profiles similar to single-agent docetaxel. Proprietary formulations such as DN-101 have been developed to optimize calcitriol delivery in this context[2][3][5][6].
