Overview

Calcitriol + gemcitabine is a combination of two pharmaceutical agents investigated primarily for their synergistic anti-tumor effects in cancer therapy. Calcitriol is the hormonally active form of vitamin D3 and acts as an agonist at the vitamin D receptor (VDR), modulating calcium homeostasis and exerting immunomodulatory and antiproliferative effects. Gemcitabine is a nucleoside analog chemotherapeutic agent that inhibits DNA synthesis by incorporating into DNA during replication and arresting tumor cell growth. Preclinical studies have shown that calcitriol can enhance the anti-tumor activity of gemcitabine by promoting caspase-dependent apoptosis in cancer cells such as pancreatic carcinoma models[3]. This combination has been explored in clinical trials for advanced pancreatic cancer[4].
