This is a combination of three medications that has been studied for potential use in prostate cancer treatment. Calcitriol (1,25-dihydroxyvitamin D3) is the active form of vitamin D that has demonstrated anticancer properties. Ketoconazole is an antifungal medication that inhibits P450 enzymes, including those involved in androgen biosynthesis and vitamin D metabolism. Hydrocortisone is a corticosteroid with anti-inflammatory properties. In preclinical studies, the combination of ketoconazole and calcitriol has shown enhanced antitumor activity against prostate cancer cells, with ketoconazole potentiating the growth inhibitory effects of calcitriol by partially blocking the enzyme that converts active vitamin D compounds to inactive products.
