Calcium carbonate + fluoride is an investigational combination regimen being evaluated for the prevention of dental caries. The treatment protocol involves the sequential administration of a chewable calcium carbonate supplement followed by a sodium fluoride mouth rinse. This approach is designed to increase the concentration and retention of fluoride in oral fluids by pre-loading the oral environment with calcium ions. Mechanistically, the elevated levels of calcium and fluoride ions in the mouth support tooth enamel mineral homeostasis by promoting remineralization and inhibiting demineralization. This strategy aims to amplify the anticaries benefit of standard fluoride treatments and is currently being studied in early Phase 1 clinical trials led by the University of Michigan.
