Drug intelligence / Profile preview
CAM-H2 (also known as [131I]-labeled anti-HER2 single-domain antibody fragment or 131I-GMIB-Anti-HER2-VHH1) is a precision radiopharmaceutical developed by Precirix (formerly Camel-IDS), a spin-off from the Vrije Universiteit Brussel (VUB). It consists of a camelid-derived single-domain antibody (sdAb/nanobody), designated 2Rs15d, that specifically targets Domain I of the human epidermal growth factor receptor 2 (HER2). The sdAb is covalently linked to the beta- and gamma-emitting radioisotope iodine-131 (131I) via a residualizing linker, N-succinimidyl 4-guanidinomethyl-3-iodobenzoate (SGMIB). This theranostic agent is designed for both targeted radionuclide therapy and SPECT imaging of HER2-overexpressing cancers, including breast, gastric, and gastroesophageal junction (GEJ) cancers, particularly in patients with brain metastases or those who have progressed on standard anti-HER2 therapies like trastuzumab. Clinical development of CAM-H2 was discontinued following the termination of its Phase I/II trial (NCT04467515) in 2024.
