Overview

Camizestrant (AZD9833) is an oral selective estrogen receptor degrader (SERD) developed to inhibit estrogen receptor signaling in hormone receptor-positive breast cancer. It works by binding to and degrading the estrogen receptor, thereby blocking the growth-promoting effects of estrogen on breast cancer cells[3][8]. Capivasertib (AZD5363, brand name Truqap) is a small molecule inhibitor of all three isoforms of AKT serine/threonine kinase (AKT1, AKT2, AKT3), a key component in the PI3K/AKT pathway frequently altered in breast cancer. By inhibiting AKT activity, capivasertib disrupts downstream signaling that promotes tumor cell survival and proliferation[1][5][6]. The combination of camizestrant and capivasertib is being investigated for advanced or metastatic hormone receptor-positive/HER2-negative breast cancer with specific genetic alterations such as PIK3CA mutations or PTEN loss[1][5].
