Overview

Camonsertib + gemcitabine is a combination therapy under clinical investigation for advanced solid tumors, particularly those with mutations sensitizing to ATR inhibition. Camonsertib (RP-3500) is an oral, potent, and selective inhibitor of the serine/threonine-protein kinase ATR, a key regulator of the DNA damage response (DDR). Gemcitabine is a nucleoside analog that inhibits DNA synthesis, resulting in cell death, most commonly used as chemotherapeutic agent in various cancers. The rationale for combining these drugs is based on synergy: camonsertib impairs the DNA repair response via ATR inhibition, sensitizing tumor cells to DNA-damaging agents such as gemcitabine. This combination is being examined primarily in patients with advanced solid tumors harboring ATR inhibitor-sensitizing mutations, with promising anti-tumor activity and molecular responses observed, especially in gynecologic malignancies[3][4][9][10]. Development is led by Repare Therapeutics, which is evaluating safety, pharmacokinetics, pharmacodynamics, and preliminary efficacy in ongoing Phase 1 clinical trials.
