Overview

This combination therapy consists of camrelizumab (a PD-1 checkpoint inhibitor), fluzoparib (a PARP inhibitor), and nab-paclitaxel (a microtubule inhibitor). It is being investigated as a neoadjuvant treatment for patients with HER2-negative breast cancer who harbor mutations in homologous recombination repair (HRR) genes, such as BRCA1, BRCA2, and PALB2. The rationale behind this triple combination is to exploit the DNA damage caused by the PARP inhibitor and chemotherapy to enhance the immune response triggered by the PD-1 inhibitor. Fluzoparib inhibits poly (ADP-ribose) polymerase, leading to double-strand DNA breaks in HRR-deficient cells, while nab-paclitaxel provides cytotoxic effects and camrelizumab prevents immune evasion by blocking the PD-1/PD-L1 pathway. This specific regimen is being evaluated in a Phase II clinical trial (NCT05761470) led by Ying Lin.
