Drug intelligence / Profile preview
This is a combination regimen comprising **camrelizumab** (a humanized anti-PD-1 monoclonal antibody), **gemcitabine** (a nucleoside analog chemotherapy agent), **capecitabine** (an oral prodrug of 5-fluorouracil), and **docetaxel** (a taxane-class cytotoxic chemotherapy drug). Each component acts via a distinct mechanism: - **Camrelizumab** blocks the interaction of PD-1 with its ligands, enhancing T-cell–mediated immune responses against tumor cells. - **Gemcitabine** incorporates into DNA as a pyrimidine antimetabolite, inhibiting DNA synthesis and inducing apoptosis. - **Capecitabine** is converted to 5-fluorouracil, which inhibits thymidylate synthase, disrupting DNA synthesis. - **Docetaxel** promotes tubulin stabilization and inhibits microtubule depolymerization, disrupting cell division and promoting apoptosis. This combination aims to utilize immune checkpoint inhibition alongside multi-agent cytotoxic chemotherapy, potentially synergizing antitumor activity. It has been investigated in advanced/metastatic cancers, particularly pancreatic cancer, where combinations of gemcitabine, capecitabine, and docetaxel have demonstrated feasibility and activity. The addition of camrelizumab introduces an immuno-oncology component, although specific studies of all four drugs together are not yet widely published[6][8].
