Overview

This is a combination regimen consisting of two immune checkpoint inhibitors, camrelizumab and sintilimab, both monoclonal antibodies targeting programmed cell death protein 1 (PD-1), together with the cytotoxic chemotherapy agent gemcitabine. Camrelizumab and sintilimab block the PD-1/PD-L1 pathway, thereby enhancing T-cell mediated antitumor immunity by preventing tumor-induced immune suppression. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, leading to apoptosis in rapidly dividing cancer cells. This combination aims to synergistically enhance antitumor effects by combining immunotherapy with cytotoxic chemotherapy. While combinations of camrelizumab or sintilimab with gemcitabine (and other agents) are under clinical investigation for various cancers such as non-small cell lung cancer (NSCLC), nasopharyngeal carcinoma, biliary tract cancer, and others in China[1][4][5][6][7], there is no evidence that this exact triple combination has been widely studied or approved as a standard regimen.
