Camrelizumab + oxaliplatin + S-1 + trastuzumab is a combination chemo-immunotherapy regimen currently being evaluated in Phase II clinical trials for the neoadjuvant treatment of HER2-positive, resectable, locally advanced gastric adenocarcinoma and gastroesophageal junction (GEJ) adenocarcinoma. This multi-modal regimen combines camrelizumab, a PD-1 checkpoint inhibitor, with trastuzumab, a monoclonal antibody targeting the HER2 receptor, and the SOX chemotherapy backbone (consisting of oxaliplatin and the oral fluoropyrimidine S-1). The combination is designed to improve pathological complete response (pCR) rates by simultaneously inhibiting immune evasion via the PD-1/PD-L1 pathway, blocking HER2-mediated oncogenic signaling, and inducing cytotoxic cell death through DNA damage and inhibition of thymidylate synthase.
