Overview

Camrelizumab + pembrolizumab is a combination of two immune checkpoint inhibitors targeting the programmed cell death protein 1 (PD-1) receptor. Both are monoclonal antibodies that block PD-1, thereby releasing PD-1 pathway-mediated inhibition of the immune response and enhancing anti-tumor immunity. Camrelizumab binds a unique glycosylated epitope (ASN58) on PD-1, while pembrolizumab binds a different epitope on PD-1; both have comparable binding affinity and receptor occupancy. This combination is used in clinical settings primarily for advanced non-small cell lung cancer (NSCLC) and other solid tumors to improve progression-free survival and overall survival compared to chemotherapy alone or monotherapy with either agent. The combination has shown manageable safety profiles in trials but is not yet established as a standard fixed-dose combined drug product; rather it represents concurrent use of two separate anti-PD-1 agents.
