This is a four-drug combination regimen consisting of camrelizumab, pemetrexed, carboplatin, and famitinib. Camrelizumab is a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), acting as an immune checkpoint inhibitor to enhance antitumor immune responses. Pemetrexed is an antifolate chemotherapy agent that inhibits multiple folate-dependent enzymes involved in nucleotide synthesis, leading to inhibition of DNA and RNA synthesis in tumor cells. Carboplatin is a platinum-based chemotherapeutic that induces DNA crosslinking and apoptosis. Famitinib is an oral small molecule tyrosine kinase inhibitor targeting vascular endothelial growth factor receptor (VEGFR) and other kinases involved in angiogenesis. The regimen has been investigated primarily as first-line therapy for advanced or metastatic non-squamous non-small-cell lung cancer (NSCLC) without EGFR or ALK alterations. Clinical trials have demonstrated improved progression-free survival and overall survival compared to chemotherapy alone, with manageable toxicity profiles[2][5][6]. The addition of famitinib aims to further inhibit tumor angiogenesis and potentially improve outcomes when combined with immunotherapy and chemotherapy[1][3].
