Camrelizumab + thalidomide is a combination therapy involving the monoclonal antibody camrelizumab and the immunomodulatory agent thalidomide. Camrelizumab is a programmed cell death protein 1 (PD-1) inhibitor used as an immune checkpoint inhibitor in cancer immunotherapy, particularly for various solid tumors. Thalidomide is a small-molecule drug with anti-inflammatory, immunomodulatory, and antiangiogenic properties. This combination has been investigated primarily to prevent or treat reactive cutaneous capillary endothelial proliferation (RCCEP), a common immune-related adverse event associated with camrelizumab therapy. Thalidomide’s mechanism includes inhibition of angiogenesis by blocking vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF), as well as modulation of inflammatory cytokines such as tumor necrosis factor-alpha (TNF-α). The rationale for combining these agents is that thalidomide can mitigate RCCEP without significantly interfering with the antitumor efficacy of camrelizumab[1][2][5].
