Overview

Canagliflozin, dapagliflozin, and empagliflozin are all sodium-glucose co-transporter 2 (SGLT2) inhibitors used as oral antihyperglycemic agents for the treatment of type 2 diabetes. Each drug lowers blood glucose by inhibiting SGLT2 in the proximal renal tubules, reducing glucose reabsorption and increasing urinary glucose excretion. They also promote mild diuresis and natriuresis. Beyond glycemic control, these agents have demonstrated cardiovascular and renal protective effects in patients with type 2 diabetes and comorbid conditions such as heart failure or chronic kidney disease. While each is approved individually or in dual combinations with other antidiabetic drugs (such as metformin), there is no evidence that a fixed-dose combination product containing all three—canagliflozin + dapagliflozin + empagliflozin—exists or is approved for clinical use[3][4][5][8]. The safety profile of SGLT2 inhibitors includes risks such as genital mycotic infections, volume depletion, hypotension, increased LDL cholesterol levels, bone fracture risk (notably with canagliflozin), and rare cases of euglycemic diabetic ketoacidosis[5][8].
