Overview

Canakinumab + docetaxel is a combination therapy consisting of canakinumab, a fully human monoclonal antibody that inhibits interleukin-1 beta (IL-1β), and docetaxel, a cytotoxic taxane-class chemotherapy agent. Canakinumab acts by blocking IL-1β-mediated inflammation, which has been implicated in cancer progression and immune evasion. Docetaxel disrupts microtubule function, inhibiting cell division and inducing apoptosis in rapidly dividing tumor cells. This combination was investigated for the treatment of advanced non-small cell lung cancer (NSCLC) in patients who had progressed after platinum-based doublet chemotherapy and immunotherapy. In the phase 3 CANOPY-2 trial, the addition of canakinumab to docetaxel did not improve overall survival compared to docetaxel alone[1][2][4].
