Overview

**canakinumab + LNA043** is an investigational combination therapy for knee osteoarthritis, combining two agents with distinct mechanisms. **Canakinumab** is a fully human monoclonal antibody that selectively neutralizes interleukin-1 beta (IL-1β), thereby reducing inflammation and pain. **LNA043** is a recombinant protein derivative of angiopoietin-like 3 (ANGPTL3), designed to induce chondrogenesis and help regenerate hyaline cartilage by acting on the integrin α5β1 (fibronectin receptor) on mesenchymal stem cells and chondrocytes. The combination is being studied to assess whether anti-inflammatory treatment with canakinumab can enhance the cartilage-regenerating effects of LNA043 in patients with inflammatory knee osteoarthritis. Both drugs are administered as intra-articular injections and are currently in clinical trials for knee osteoarthritis[1][2][3][4][5][7].
