candesartan cilexetil + amlodipine

Clodipan is a fixed-dose combination of candesartan cilexetil, an angiotensin II receptor blocker (ARB), and amlodipine, a dihydropyridine calcium channel blocker (CCB). It is approved for the treatment of hypertension (high blood pressure). Candesartan cilexetil is a prodrug that is hydrolyzed to candesartan, which selectively blocks the binding of angiotensin II to the AT1 receptor, leading to vasodilation and decreased aldosterone secretion. Amlodipine inhibits the transmembrane influx of calcium ions into vascular smooth muscle and cardiac muscle, resulting in peripheral arterial vasodilation and reduced peripheral vascular resistance. Together, these complementary mechanisms provide synergistic blood pressure reduction and organoprotective effects.
