This combination drug consists of **canerpaturev** (C-REV, HF10)—a genetically modified, attenuated oncolytic herpes simplex virus type-1—combined with **gemcitabine** (a nucleoside analog antimetabolite), and **nab-paclitaxel** (albumin-bound paclitaxel, a microtubule inhibitor). Canerpaturev selectively infects and lyses tumor cells, stimulating local and systemic antitumor immune responses, including recruitment and activation of cytotoxic T lymphocytes. Nab-paclitaxel stabilizes microtubules, preventing cell division and promoting apoptosis, while the albumin-based delivery improves tumor penetration and accumulation via interaction with tumor stroma and possibly SPARC (Secreted Protein Acidic and Rich in Cysteine)[1][2][4][6][8]. This combination is under clinical investigation for advanced pancreatic cancer, aiming to synergize oncolytic immunotherapy and chemotherapy by enhancing tumor killing, modulating the microenvironment, and depleting tumor stroma.
