Canerpaturev + ipilimumab is an investigational combination of two immunotherapies: **canerpaturev** (C-REV, formerly HF10), a naturally occurring oncolytic herpes simplex virus type 1 (HSV-1) mutant, and **ipilimumab**, a fully human monoclonal antibody that blocks cytotoxic T-lymphocyte antigen-4 (CTLA-4). Canerpaturev targets and lyses tumor cells directly as an oncolytic virus while simultaneously inducing immune responses against cancer, whereas ipilimumab augments T-cell mediated anti-tumor immunity by inhibiting the CTLA-4 checkpoint pathway on T cells. Preclinical research suggested a synergistic effect: canerpaturev enhances immune recognition, and ipilimumab further boosts anti-tumor immune activity. The combination has been evaluated in phase II clinical trials to assess safety and efficacy in patients with advanced or metastatic melanoma, with reported adverse events but promising indications of immunologic activity[5].
