Canfosfamide + carboplatin is a combination regimen investigated primarily in oncology. Canfosfamide (also known as TLK286, Telcyta) is a glutathione analog prodrug activated by glutathione S-transferase P1-1, an enzyme often over-expressed in various tumors. Once activated, canfosfamide generates cytotoxic moieties that induce apoptosis in cancer cells. Carboplatin is a platinum-based chemotherapeutic that forms DNA crosslinks, leading to apoptosis. This combination was studied especially in platinum-resistant or platinum-refractory ovarian cancer and in non-small cell lung cancer (NSCLC). The combination did not show survival benefits in phase 3 ovarian cancer trials, but demonstrated activity and acceptable safety in early phase NSCLC studies[1][2][3][4].
